Looks like a type of red turf algae. I wouldn't think an algae would "close up" however.
What brand of skimmer do you have? Can you put something under the skimmer to raise it which will lower the water level?

Some test kits are made for saltwater and may not work with fresh tap water. You may have gotten a false negative reading.
