Is there an air bubble in the top of the U-tube? Did you fill the tube with water, and then keep a thumb over both ends until you got it fully into place?

when I had the overflow box, I would stick a piece of airline tubing in the back end so that the end of the tube in the u-tube was right at the bend, and just suck on the other end of the airline tubing. After practice,I was able to do this without swallowing saltwater.
I'm sure there are other ways to do it, that's what worked best for me.

make sure both ends of the u-tube are under water esp the end in the tank. raise the tanks water level it will help. you need to get a piece of airline tubing and stick it into your u-tube and get it right to the top of the u-tube. suck the air out of the u-tube(i hope you like the taste of saltwater) and have someone kick on your return pump. the siphon should start and if you have a good enough pump no bubbles will remain in the utube.
