I took a couple of pieces of LR out of my tank to kill some aiptasia that was starting to take over, I put the rock in a bag in my basement. They have been there for like 1 week, When can I put them back in my tank and what should I do before i put them back in my tank?

It is a dry heat, yeah right !
I would rinse them well, especially since they have been stored in a moist environment. Try to get as much die off removed as you can.

Can it be with regular water or does it have to be with RO/DI water?

Rinse in change water to be disposed and scrub with a brush to remove as much die off as possible. You want to try to minimize the chance of a mini cycle.

Use salt water from a water change.
