Want to build a refugium, have some questions

Hi guys,
i want to build a refugium for my 110 gallon tank.
I am probably going to convert a 40 gallon breeder into my sump/refugium.

I have some questions though.
1. I only have one corner overflow, whats the best layout of compartments i should use. Baffles???
2. Is it better to plumb the intake and return lines with rigid PVC, flexible PVC, or regular clear lines?
3. How long does it take for the system to be working at 100%
4. How do i start the refugium, meaning get the algae growing.

and if you start with base sand - you can connect immediately since there is no die-off and major cycling associated with it. Livesand - you would want to isolate the tank and give it flow for a month before connecting.
