Right now I have one picasso clown in my 12g nano cube. I plan on getting another this week. My question is Should I put the pair in my 12g nano cube or should I take the one that is currently in my 12g and add them both at the same time into my 20g L. Im debating on which tank to put them into. I know its probally better for them to be in the bigger tank but if they have something to host they really dont stray to far from it.

you probably should add them both the same time to the new tank. Is there a size difference between the two?

I wouldn't worry about adding the other to the 12g. Those clowns are not very aggressive - I believe they're the same as true percs, just a genetic mutation.

I would add them both to a new tank. Though they are more peaceful clowns, clowns can be nasty and unpredictable when adding a new clown to an established clown's tank.
